b-Caseinephospholipid monolayers as model systems to understand lipideprotein interactions in the milk fat globule membrane

چکیده
Phospholipideprotein monolayer films were studied as model systems to mimic the structure of the native bovine milk fat globule membrane (MFGM) and to understand lipideprotein interactions at the surface of the globule. Phospholipids extracted from bovine raw milk, raw cream, processed milk and buttermilk powder were spread onto the airewater interface of a Langmuir trough, b-casein was then added to the sub-phase, and LangmuireBlodgett films were studied by epifluorescence microscopy and atomic force microscopy. In all films, b-casein was responsible for clustering of the sphingomyelinand cholesterol-rich microdomains into larger platforms. This suggests that the same phenomenon may happen at the surface of the milk fat globule, where specific MFGM proteins may cause aggregation of

Butyrophilin controls milk fat globule secretion.
The molecular mechanism underlying milk fat globule secretion in mammary epithelial cells ostensibly involves the formation of complexes between plasma membrane butyrophilin and cytosolic xanthine oxidoreductase. These complexes bind adipophilin in the phospholipid monolayer of milk secretory granules, the precursors of milk fat globules, enveloping the nascent fat globules in a layer of plasma...
متن کاملInteractions of fat globule surface proteins during concentration of whole milk in a pilot-scale multiple-effect evaporator.
The changes in milk fat globules and fat globule surface proteins during concentration of whole milk using a pilot-scale multiple-effect evaporator were examined. The effects of heat treatment of milk at 95 degrees C for 20 s, prior to evaporation, on fat globule size and the milk fat globule membrane (MFGM) proteins were also determined. In both non-preheated and preheated whole milk, the size...
متن کاملFractionation of globular milk fat by membrane microfiltration
Separation of milk fat in small globules (SG) (diameter lower than 2 μm) and in large globules (LG) (diameter higher than 2 μm) was realized by a patented process using special ceramic microfiltration membranes. Transformations in drinking milks, yoghourts, sour cream, camembert, Swiss cheese and butters were realized from milks of which the fat content was adjusted either by reference cream or...
متن کاملThe endoplasmic reticulum and casein-containing vesicles contribute to milk fat globule membrane
During lactation, mammary epithelial cells secrete huge amounts of milk from their apical side. The current view is that caseins are secreted by exocytosis, whereas milk fat globules are released by budding, enwrapped by the plasma membrane. Owing to the number and large size of milk fat globules, the membrane surface needed for their release might exceed that of the apical plasma membrane. A l...
متن کاملRe-evaluation of milk-fat globule EGF-factor VIII (MFG-E8) as an intrinsic component of the mouse milk-fat globule membrane.
In lactating mammary glands, milk fat is secreted as fat globules surrounded by a cell plasma membrane containing characteristic membrane-associated proteins. Among these, butyrophilin has been shown to be specific and intrinsic to the fat globule membrane, whereas milk-fat globule EGF-factor VIII (MFG-E8) is uncertain. We characterized in the present study MFG-E8 in milk fat globules and in th...
